OpenGL or DirectX -- I don't think it's a detriment to learn one or the other; you're going to have to be trained in some areas, and if you're good enough to know one, the other probably won't be too huge of a deal to become familiar with.
I just prefer OpenGL because it's cross-platform, and not a Microsoft product. :p
